I am sure the answer to this is blindingly easy, and I'll gladly kick myself when I get it...
Existing Exchange 2007 environment:
VMPCS021 - 2007 SP3 Edge on 2008 R2
VMPCS110 - 2007 SP3 CAS/HUB/Mailbox on 2008 R2 - Running public folders
VMPCS111 - 2007 SP3 CAS/HUB/OWA on 2008 R2
Added new Exchange 2010 SP1 servers:
VMPCS112 - 2010 SP1 CAS/HUB/Mailbox on 2008 R2 - running public folders. In DAG with 113
VMPCS113 - 2010 SP1 CAS/HUB/Mailbox on 2008 R2 - running OWA. In DAG with 112
VMPCS115 - 2010 SP1 CAS/HUB on 2008 R2 - Witness server for DAG
My situation is:
Mailboxes on 2010 can send mail to mailboxes on 2007 or 2010 exchange servers. They are not able to send email to the internet.
Mailboxes on 2007 can send mail to 2007 mailboxes but not 2010. They can send and recieve from the internet.
I removed the edge server subscription on vmpcs021 and created a new one and imported it in to replace the old certificate. When I try and send mail from 2007 mailboxes to 2010 mailboxes, it goes into the queue on vmpcs110 and eventually dies as 550 4.4.37 QUEUE.Expired.
If I try and send to an external address it eventually dies with the same error. I have pasted the NDR below:

What gives? Any ideas on this would help, and as I said, I figure it's something blindingly simple. I have removed and reinstalled both the hub and cas roles on both vmpcs112 and vmpcs113 with no change in the behavior.
